Do these grab rails interfere with the saddlebags being opened on a Road King Classic?

It depends on what model year machine it is that you have. For 2008 and older FLHRC models, you can still open the saddlebags, but just not all the way. And that is a bit annoying! For the 2009+ FLHRC, you can open the bags all the way without issue.
