Will this sprocket work with a o ring chain

Yes, this will work with an O-Ring chain. The chain type of chain you have does not matter when it comes to sprockets. What's most important is that you're matching chain size (called chain pitch) to the correct sprocket. If you've got a 520 chain, be it unsealed, Z-Ring, X-Ring &etc., it will work well with this sprocket.
How would I know if my 2008 Yamaha wr250r has a "520" chain?

That 2008 Yamaha WR250R would have come from the factory with a 520 pitch chain/sprockets. With that said, you should have no problem using this Sunstar 520 sprocket.
